Various types of myogenic stem cells (SC), including muscle SKs, that can be isolated from skeletal muscle as a population of cells slowly attached to the collagen-coated surface were used to repair the damaged muscle or muscle tissue injury. The therapeutic use of muscle stem cells to improve muscle regeneration seems very promising, but the effect of tissue damage on the characteristics and survival of these cells has not yet been studied, according to an online edition for girls and women aged 14 to 35 Pannochka. Net American scientists from the University of Pittsburgh isolated and studied a population of cells slowly attached to a slow-adhering stem cells (SASCs) from damaged and intact (control) tissue of the skeletal muscles of mice. The in vitro rate of migration and proliferation, the ability to multipotent differentiation, the differences in the gene expression of these two groups of cells.

It has been shown that a much larger population of SASCs can be isolated from the injured muscle than from an intact. In addition, these cells were more likely to migrate, proliferated at a much higher rate and had increased expression of Notch1, STAT3, Msx1, and MMP2.

In the next stage of the experiments, SASCs were transplanted into the muscles of MDX / SCID muscular dystrophy mice.

It was found that the cells obtained from the injured muscle were much better established and helped to form a denser capillary network than cells from intact musculature.

Thus, the data obtained in the study indicate that damage to muscle tissue changes the characteristics of muscle stem cells. Apparently, this effect is mediated by trophic factors. The results of the study can be used to increase the effectiveness of cellular therapy for muscle damage.
