Improved method of cell transplantation to restore the retina

The inability of an adult mammalian retina to regenerate is in part related to the barrier resulting from the retinal degeneration of the inhibitory extracellular matrix (ECM) and cell adhesion molecules such as CD44 and neurocan (neurocan).

These molecules do not allow the subretinal graft to integrate with the retina. It has been shown that matrix metalloproteinases-2 (MMP2), by destroying these molecules, can facilitate integration of the graft.

Chinese scientists from Fudan University performed a series of experiments on retinal cell therapy in adult Rho - / - mice that have genetically determined retinal degeneration. To enhance cellular integration and stimulation of retinal repopulation, in addition to the transplantable retinal progenitor cells (RPCs), microspheres that release active MMP2 are introduced into the subretinal space of the PLGA (lactic-co-glycolic acid).

There was destruction of CD44 and neurikana on the outer surface of the degrading retina without disturbing its architectonics. Simultaneously, the number of cells migrating through the barrier to the degenerating retina significantly increased, and no changes in the differential characteristics of RPCs occurred. The cells of the outer nuclear layer expressed the markers of mature photoreceptors (receptorin), established contacts with protein kinase positive cells C and expressed the synaptic bassoon protein.



Thus, the introduction of the RPCs into the degradation retina, together with the PLGA microspheres releasing active MMP2, removes the inhibitory barrier and improves the integration of the transplanted cells. According to Dr. P. Katunyan, the head physician of the Moscow Center for Biomedical Technologies, in case of the success of the relevant clinical trials, this method can be recommended for clinical use as an effective retinal recovery strategy.
