Donation of blood stem cells is safe

Currently, the main source of stem cells for transplantation is not the bone marrow, as it was before, but peripheral blood. Therefore, the use of the G-CSF cytokine, which mobilizes stem cells from bone marrow to peripheral blood. There were fears that even a short-term exposure to low doses of G-CSF on healthy donors could have a genotoxic effect.

American scientists from the University of Minnesota Medical School used the FISH (fluorescence-in-situ-hybridization) method to study peripheral blood lymphocytes to assess the degree of chromosomal instability. During the year five lymphocytes of 22 donors of peripheral blood stem cells and 22 of the control group not exposed to the cytokine.

Blood donors were taken for analysis prior to G-CSF administration, 5-7 days after administration and at 2, 6 and 12 months. Another 8 blood donors received blood once every 12 months after exposure to a cytokine.

On aneuploidy, nine loci were studied in chromosomes 7, 8, 9, 17, 21 and 22, including three loci of chromosome 7, since special fears caused monosomy along this chromosome. The time of replication at the loci of chromosomes 15 and 17.

There was no evidence of chromosomal instability caused by the use of G-CSF.

Thus, the results of this paper are consistent with those obtained in epidemiological studies that also showed no risk of hematological malignancies for peripheral blood stem cell donors who received G-CSF injections.
